The droplet problem

The limit is often not the molecule. It is the droplet.

Many valuable active ingredients, such as cannabinoids, carotenoids, CoQ10 and fat-soluble vitamins, are lipophilic. How they are presented to the gut, the skin, or a beverage depends heavily on the size and uniformity of the oil droplets that carry them.

≈6%

Estimated oral bioavailability of cannabidiol (CBD) in fasted humans.

Perucca & Bialer, CNS Drugs, 2020

a = 6 / d

Interfacial area

For a fixed oil volume, taking droplets from 1 µm to 50 nm multiplies the oil–water interface twentyfold, which speeds lipolysis and micellar solubilization.

I ∝ d⁶ / λ⁴

Optical clarity

Well below the wavelength of light, scattering falls with the sixth power of diameter. Halve the droplet and scattering drops about 64-fold, so the emulsion turns clear.

PDI ≤ 0.3

Stability

A narrow size distribution slows Ostwald ripening. Below 100 nm, Brownian motion outweighs gravity, so creaming slows dramatically.

Process data · BaroShear NE-5

135 nm to 46 nm in six passes.

Starting emulsion shown pre-processing, and then after each pass through the BaroShear NE-5. Mean droplet diameter falls steeply over the first few passes and then begins to level off, and the sample clarifies as it does. Pass count is the simplest lever you have.

How UltraShear works

One patented valve. Three levers you control.

A coarse starting emulsion is pressurized to as much as 30,000 psi and forced through an annular valve whose gap is in the nanometer range. The valve is self-throttling: its clearance adapts dynamically, which is designed to resist the clogging and erosion that limit fixed-orifice valves.

Peer-reviewed evidence

The first in-vivo data, with its limits.

ElSohly, Shahzadi and Gul (2023) dosed a ≈60 nm UltraShear CBD nanoemulsion to rats and compared bioavailability with a dose-adjusted IV reference. The formulation was made on the BaroShear Mini benchtop system.

Relative to dose-adjusted IV (AUC ratio)
F · parent CBD · 6 h18.6%
F · parent CBD · 24 h25.4%
A · CBD-equivalents · 6 h45.5%
A · CBD-equivalents · 24 h79.7%

F = parent CBD; A = CBD plus 7-OH-CBD and 7-COOH-CBD as CBD-equivalents. Oral n = 4, IV n = 3. Open-access paper (PMC10629855). The study was partly funded by Pressure BioSciences.

What a reviewer will ask

  • Very small groups: four rats orally and three by IV.
  • No matched oral control in the same study, so the size of any advantage over a conventional formulation is not yet measured.
  • The IV arm used a different vehicle, so route and vehicle are confounded.
  • Rat data do not translate directly to people. Matched-control and human PK studies are the next step.

Applications

Where teams evaluate UltraShear.

The physics applies to any lipophilic active ingredient carried in an oil phase. Each product should be judged on its own data; these are candidate applications, not performance claims.

Pharmaceutical R&D

Poorly water-soluble APIs and lipid-based oral or topical systems that need small, narrow droplets and sterile filtration where possible.

Nutraceuticals

Oil-soluble active ingredients where higher bioavailability is the goal, such as carotenoids, CoQ10, curcumin, omega-3, astaxanthin, and vitamins A, D, E and K in clear, stable liquid formats with less surfactant.

Cannabinoids and botanicals

CBD, THC, minor cannabinoids, and botanical extracts for water-compatible beverages, sprays, and drops designed for fast onset.

Food and beverage

Flavor oils, colors, and fortificants in transparent beverages, processed cold.

Cosmetics and personal care

Oils, vitamins, and botanical active ingredients in light, clear serums and sprays.

Agrochemical and industrial

Oil-soluble actives and adjuvants in concentrates that stay stable on dilution.

Questions

Before you apply.

Anything else, ask Ric directly: rschumacher@pressurebiosciences.com.

What is the BaroShear NE-5 Gen 2?

A medium-sized, floor-model ultra-high-pressure homogenizer for oil-in-water nanoemulsions on the patented UltraShear Technology (UST) platform. It runs at 10,000–30,000 psi with continuous flow of up to 0.8 L/min per pass, for R&D, pilot and manufacturing runs.

What droplet sizes can it reach?

Final droplets range from above 200 nm down to about 20 nm, set by your formulation, the pressure, and the number of passes. In Pressure BioSciences' own data, one emulsion went from 135 nm to 46 nm in six passes on the NE-5.

Why are build slots limited?

The first production run has a limited number of systems, so Pressure BioSciences allocates them by application, after a conversation with each team about its formulation and site. Applying is free and carries no obligation.

How do I get pricing and availability?

Send the short application form. Ric Schumacher or one of our scientists replies within one business day and provides pricing, availability and purchase terms in writing.

Do I own the machine, or lease it?

You own it outright as your own capital asset, together with a right to use the UltraShear patents under Pressure BioSciences' license terms. There is no lease and nothing to hand back. Ric explains the full purchase and license terms in writing after you apply.

Is financing available?

Yes, for qualified non-cannabis buyers, with no payments for the first four months. Financing requires bank approval and a refundable security deposit. Ask Ric for terms; he provides them in writing after you apply.

Can cannabis companies buy or finance the NE-5?

Cannabis and psychotropic producers can buy the NE-5, but bank financing through Pressure BioSciences is not available for that use. For cannabinoids and psychotropics the machine is for your own production only, with no contract processing for others, and it stays in the state where it is installed.

Can I co-brand my product with UltraShear?

Eligible customers may label products "Made with UltraShear", subject to Pressure BioSciences' approval and a written agreement. We also help with formulation so the product earns the mark. Tick "Co-branding" when you apply.

Can you process my formulation first?

It is sometimes possible to process your formulation and report particle size, PDI and appearance by pass. There may be a reasonable charge for this service.

What does my site need?

208–240 V AC, 1-phase, 50/60 Hz, 30 A power and at least 8 × 8 ft of operating space. DI and potable water, drainage, filtration cassettes and consumables are provided by the customer; we advise on specifications.

What happens after the warranty year?

Year one is covered. From year two an optional extended service contract is available for one, two or three years; Ric includes the terms with your purchase terms.

What is included, and what is the warranty?

Two 10 L jacketed tanks, stainless-steel heat-exchange coils with a chiller, and a stainless-steel enclosure rated IP-54 or better. The warranty is one year on non-consumable parts, travel and labor.

What is the evidence on bioavailability?

One peer-reviewed rat study (ElSohly et al., 2023) with small groups and no matched oral control. Matched-control and human PK studies are the next step. This page makes no therapeutic or health claims.
